RNA-based therapy shows promise for treating genetic diseases caused by nonsense mutations

A new therapeutic strategy based on RNA has been developed that could potentially treat a wide range of genetic diseases caused by nonsense mutations, such as cystic fibrosis and some forms of muscular dystrophy. The research, published in Science, was conducted by scientists at the University of Toronto in Canada. The approach uses suppressor tRNAs (sup-tRNAs) that recognize premature stop signals in messenger RNA and insert an amino acid to allow continued protein synthesis, producing a full-length, potentially functional protein. To overcome previous limitations of suboptimal activity and inefficient delivery, the researchers chemically modified the sup-tRNAs and packaged them in lipid nanoparticles (similar to those used in COVID vaccines) for inhalation delivery to the lungs. The strategy proved effective in initial tests on bronchial epithelial cells, laboratory mice, and organoids derived from cystic fibrosis patients. The long-term goal is to develop tRNA-based drugs that recognize common stop signals, potentially applying a single therapeutic strategy to many different genetic diseases.
